    NASDA's Advanced On-Line System (ADOLIS)

    Spacecraft operations including ground system operations are generally realized by various large or small scale group work which is done by operators, engineers, managers, users and so on, and their positions are geographically distributed in many cases. In face-to-face work environments, it is easy for them to understand each other. However, in distributed work environments which need communication media, if only using audio, they become estranged from each other and lose interest in and continuity of work. It is an obstacle to smooth operation of spacecraft. NASDA has developed an experimental model of a new real-time operation control system called 'ADOLIS' (ADvanced On-Line System) adopted to such a distributed environment using a multi-media system dealing with character, figure, image, handwriting, video and audio information which is accommodated to operation systems of a wide range including spacecraft and ground systems.     Mechanical loss of a multilayer tantala/silica coating on a sapphire disk at cryogenic temperatures: toward the KAGRA gravitational wave detector

    We report the results of a new experimental setup to measure the mechanical loss of coating layers on a thin sapphire disk at cryogenic temperatures. Some of the authors previously reported that there was no temperature dependence of the mechanical loss from a multilayer tantala/silica coating on a sapphire disk, both before and after heat treatment, although some reports indicate that Ta<sub>2</sub>O<sub>5</sub> and SiO<sub>2</sub> layers annealed at 600 °C have loss peaks near 20 K. Since KAGRA—the Japanese gravitational-wave detector, currently under construction—will be operated at 20 K and have coated sapphire mirrors, it is very important to clarify the mechanical loss behavior of tantala/silica coatings around this temperature. We carefully investigate a tantala/silica-coated sapphire disk with the new setup, anneal the disk, and then investigate the annealed disk. We find that there is no distinct loss peak both before and after annealing under particular conditions. The mechanical loss for the unannealed disk at 20 K is about 5×10<sup>−4</sup>, as previously reported, while that for the annealed disk is approximately 6.4×10<sup>−4</sup>

    Lattice-patterned collagen fibers and their dynamics in axolotl skin regeneration

    The morphology of collagen-producing cells and the structure of produced collagen in the dermis have not been well-described. This lack of insights has been a serious obstacle in the evaluation of skin regeneration. We succeeded in visualizing collagen-producing cells and produced collagen using the axolotl skin, which is highly transparent. The visualized dermal collagen had a lattice-like structure. The collagen-producing fibroblasts consistently possessed the lattice-patterned filopodia along with the lattice-patterned collagen network. The dynamics of this lattice-like structure were also verified in the skin regeneration process of axolotls, and it was found that the correct lattice-like structure was not reorganized after simple skin wounding but was reorganized in the presence of nerves. These findings are not only fundamental insights in dermatology but also valuable insights into the mechanism of skin regeneration

    Advanced Tertiary Lymphoid Tissues in Protocol Biopsies are Associated with Progressive Graft Dysfunction in Kidney Transplant Recipients

    移植腎の三次リンパ組織は腎予後と関連することを解明. 京都大学プレスリリース. 2021-11-02.[Background:] Tertiary lymphoid tissues (TLTs) are ectopic lymphoid tissues found in chronically inflamed organs. Although studies have documented TLT formation in transplanted kidneys, the clinical relevance of these TLTs remains controversial. We examined the impacts of TLTs on future graft function using our histological TLT maturity stages and the association between TLTs and Banff pathologic scores. We also analyzed the risk factors for the development of TLTs. [Methods:] Serial protocol biopsy samples (0-hour, 1-, 6-, and 12-months) without rejection were retrospectively analyzed from 214 patients who underwent living donor kidney transplantation. TLTs were defined as lymphocyte aggregates with signs of proliferation and their stages were determined by the absence (stage I) or presence (stage II) of follicular dendritic cells. [Results:] Only 4% of patients exhibited TLTs at the 0-hour biopsy. Prevalence increased to almost 50% at the 1-month biopsy and then slightly further for 12 months. The proportion of advanced stage II TLTs increased gradually, reaching 19% at the 12-month biopsy. Presence of stage II TLTs was associated with higher risk of renal function decline after transplantation compared to patients with no TLT or stage I TLTs. Stage II TLTs were associated with more severe tubulitis and interstitial fibrosis/tubular atrophy at 12 months and predicted poorer graft function independently from the degree of interstitial inflammation. Pre-transplantation rituximab treatment dramatically attenuated the development of stage II TLTs. [Conclusions:] TLTs are commonly found in clinically stable transplanted kidneys. Advanced stage II TLTs are associated with progressive graft dysfunction, independent of interstitial inflammation

    Heme Orientation of Cavity Mutant Hemoglobins (His F8 → Gly) in Either α or β Subunits: Circular Dichroism, 1H NMR, and Resonance Raman Studies

    Native human adult hemoglobin (Hb A) has mostly normal orientation of heme, whereas recombinant Hb A (rHb A) expressed in E. coli contains both normal and reversed orientations of heme. Hb A with the normal heme exhibits positive circular dichroism (CD) bands at both the Soret and 260-nm regions, while rHb A with the reversed heme shows a negative Soret and decreased 260-nm CD bands. In order to examine involvement of the proximal histidine (His F8) of either α or β subunits in determining the heme orientation, we prepared two cavity mutant Hbs, rHb(αH87G) and rHb(βH92G), with substitution of glycine for His F8 in the presence of imidazole. CD spectra of both cavity mutant Hbs did not show a negative Soret band, but instead exhibited positive bands with strong intensity at the both Soret and 260-nm regions, suggesting that the reversed heme scarcely exists in the cavity mutant Hbs. We confirmed by 1H NMR and resonance Raman (RR) spectroscopies that the cavity mutant Hbs have mainly the normal heme orientation in both the mutated and native subunits. These results indicate that the heme Fe-His F8 linkage in both α and β subunits influences the heme orientation, and that the heme orientation of one type of subunit is related to the heme orientation of the complementary subunits to be the same. The present study showed that CD and RR spectroscopies also provided powerful tools for the examination of the heme rotational disorder of Hb A, in addition to the usual 1H NMR technique.     Construction of scales measuring educational effects on the "Power to live".

    近年、学校教育において、生きる力の育成が目標とされている。本研究は生きる力の3要素である確かな学力、豊かな心、及び健やかな体に、信頼される校区・学校を加えた4領域の教育効果を測定する質問紙を作成するとともに、4領域の相互関係について分析することを目的とする。教育実践者への聞き取り調査から得られた項目から予備質問紙を作成し、調査後の結果に対して、領域毎に因子分析を適用した。各領域とも1因子構造が示され、因子負荷量の高い項目を参考に各領域の因子名を「確かな学びの形成力」「自尊感情」「健康志向性」「学校肯定感」とし、4尺度を構成した。「自尊感情」と他3尺度の関連性を検討したところ、「自尊感情」は他の3尺度との間に強い関連性を示したことから、自尊感情は生きる力の形成に大きな影響を与えることが示唆された。Developing the "Power to Live" has become an objective in recent education. The objectives of this study was to construct a scale to measure the educational effects of the three domains of the "Power to Live": "evident academic ability", "enriched mind", "healthy body" in addition to "reliable school and district", and to analyze the interactions among these four domains. The pre-questionnaire was constructed by items extracted from interviews with school teachers. Factor analysis was used on the results of the pre-questionnaire survey and demonstrated a mono-factor structure in each domain. According to factor loadings in items, the following factors were designated; "ability to form reliable learning"; "feeling of self-esteem", "orientation for health" and "positive feelings for school". "Feeling of self-esteem" was strongly related to the three other scales. These results showed that feelings of self-esteem influence the "Power to Live"
